Meiotic Spindle Self-Organization: One Plus One Equals Only One

چکیده
Like other cellular organelles, the bipolar spindle is a structure of well-defined size and shape. Now, spindle fusion experiments using micro-manipulation have allowed current models for spindle morphogenesis and size control to be tested.

Self-organization of stabilized microtubules by both spindle and midzone mechanisms in Xenopus egg cytosol
Previous study of self-organization of Taxol-stabilized microtubules into asters in Xenopus meiotic extracts revealed motor-dependent organizational mechanisms in the spindle. We revisit this approach using clarified cytosol with glycogen added back to supply energy and reducing equivalents. We added probes for NUMA and Aurora B to reveal microtubule polarity. Taxol and dimethyl sulfoxide promo...
متن کاملSimulated Microgravity Compromises Mouse Oocyte Maturation by Disrupting Meiotic Spindle Organization and Inducing Cytoplasmic Blebbing
In the present study, we discovered that mouse oocyte maturation was inhibited by simulated microgravity via disturbing spindle organization. We cultured mouse oocytes under microgravity condition simulated by NASA's rotary cell culture system, examined the maturation rate and observed the spindle morphology (organization of cytoskeleton) during the mouse oocytes meiotic maturation. While the r...
متن کاملZYG-9, A Caenorhabditis elegans Protein Required for Microtubule Organization and Function, Is a Component of Meiotic and Mitotic Spindle Poles
We describe the molecular characterization of zyg-9, a maternally acting gene essential for microtubule organization and function in early Caenorhabditis elegans embryos. Defects in zyg-9 mutants suggest that the zyg-9 product functions in the organization of the meiotic spindle and the formation of long microtubules. One-cell zyg-9 embryos exhibit both meiotic and mitotic spindle defects. Meio...
متن کاملAxin-1 Regulates Meiotic Spindle Organization in Mouse Oocytes
Axin-1, a negative regulator of Wnt signaling, is a versatile scaffold protein involved in centrosome separation and spindle assembly in mitosis, but its function in mammalian oogenesis remains unknown. Here we examined the localization and function of Axin-1 during meiotic maturation in mouse oocytes. Immunofluorescence analysis showed that Axin-1 was localized around the spindle. Knockdown of...
متن کاملA One-Sided View of Kinetochore Attachment in Meiosis
Meiosis includes a reductional division in which homologous chromosomes, rather than sister chromatids, are segregated to opposite poles of the spindle. In this issue of Cell, report that casein kinase 1 contributes to this process by promoting the attachment of both kinetochores of a homolog to only one pole of the meiotic spindle in budding yeast.
